Abstract : Replacement, reduction and refinement (the 3R's) are the leading principles in translational research with animals. To be useful a model should also be clinically Relevant (the 4th R). Work in a non‐human primate model of multiple sclerosis, the experimental autoimmune encephalomyelitis model, reveals an inherent conflict among these 4R principles. The impossibility to harmonize all 4R's forms a major challenge when the model is applied in preclinical drug development. Immune disorders: Preferring primate models: Using non‐human primates in model systems could improve the chance of developing treatments for immune disorders. Current preclinical research mainly uses mice and is poor at predicting clinical success. Laboratory mice are bred in very clean conditions which limit the maturation of their immune system. Bert't Hart of the Biomedical Primate Research Centre, Rijswijk, The Netherlands suggests that reviewers should prioritize the relevance of the chosen animal model when evaluating publications and project proposals. He argues that non‐human primates such as marmosets should be used in order to make the findings clinically relevant. While smaller sample sizes would reduce the statistical significance of studies, the genetic heterogeneity and mature immune system of non‐human primates make these animals a more relevant model, increasing the likelihood that the findings will translate into treatments.
